Tissue Growth Promoting and Inhibiting Factors in Body Fluid


Abstract
Many workers 17) have carried out experiments on growth Promoting 1177 and growth Inhibiting 18261 substances extracted from various normal and tumor tissues.
Malmgren & Mills; (1961) 30), Stich & FIorian (1948) 29i, Lee & Lee (1962) s> Herbut et al 3133¢¥ reported the effects of factors on normal serum in normal tissues.
Lee 2, also reported that the effects of tissue factors extracted from beef peritoteum on mitosis of liver and kidney of normal mice.
The purpose of present work is to studyt,the growth inhibiting and promoting substances from body fluids as a part of our project on various tissue factors and to observe the effects of these factors on mitosis of liver and kidney of normal mice.
Materials and Methods
Urine from normal human, ascites from liver cirrhosis patient, bile from fistula of ccrrrr-cn bile duct and pus from pyothorax patient were used as materials.
The fresh, materials were centrifuged at 15, 000 r.p.m. for 40 minutes in an International Centrifuge Model PRI.
One volume of 95% ethylalcohol was added to same volume of the supernatant then left overnight at 40C for 24 hours and again centrifuged at 1,500 r.p.m. for 20 minutes.
The precipitate (HAP) and supernatant (HAS) thus obtained were separately dried and made a solution with saline into 5 % or 2.5 % solution. Daily dosis of fluid factor for injection was 0.2 cc of 5 % solution, except 2.5 % of bile inhibiting factor.
Young, healthy mice and same aged, weighing 1719 gm were selected an experimental animals. Special attention was paid in order to feed them under the same nutritional and environmental conditions.
One group of experimental mouse was number of 30 mice. Body fluid factor was injected into peritoneum at th e same hour daily for consecutive days. Control groups received a corresponding amount of normal saline intraperitoneally.
The groups injected daily were sacrificed at the same hour every third days following the initial injection. Four hours before slaughter, 0.01 % colchicine solution was injected intraperitoneally in order to facilitate the mitotic cell couns. Specimens were taken from a certain part of the liver and kidney, fined in Carnoy¢¥s solution for 3 hours, treated with alcohol and xylol, embedded in paraffin. cut in sections 5 microns thick, and
stained with hematoxylin and eosin. The mitotic cell counts were made in 100 fields on each slide with the help of 43 x objective lens and 10 x ocular lens.
Result
1) Growth inhibiting factor and promoting factor may be extracted from various body fluid.
2) Promoting factor enhances the mitosis of normal tissue in the early stage of injection, and reaches to
maximum, then its number is decreased and gradually approached to the number of control despite of consecutive injection.
3) Inhibiting factor curbs the mitosis of P.ormal tissue in the early stage of injection and reaches to minimum, then its number is conversely increased to near the control.
4) Intensit- of action in both factors is different according to the body fluid.
